First layer - Prepare cake mix as directed on box. Bake cake in a 11x9 inch pan or use 2 mixes and bake in an 11 x 15 inch pan for a larger cake.
Second layer - In a saucepan, bring granulated sugar and 1/2 cup milk to a boil. Add the peanut butter and marshmallow crème. Mix well. Quickly pour on top of cooled cake and spread evenly. Allow to cool.
Third layer - In a saucepan melt butter, cocoa and 1/3 cup milk. Bring to a boil. Remove from heat and add confectioners sugar and vanilla. Stir well. Spread on top of cake. Allow to cool completely before serving.
